2. Materials and methods
2.1. Qualitative screening of phytase producing bacterial strains
Samples for the isolation of phytase producers were collected
from soil attached to the root nodules of been, for primary
screening, one ml of soil suspensions was plated onto phytase
screening turbid agar media plates containing: 0.5% Caphytate
and 2% agar and incubated at 28 C. Fungal isolates
capable of hydrolyzing Ca-phytate recognized by their
surrounding clear halo zone were selected and repeatedly
streaked on potato dextrose agar slants.
