Lipid oxidation
Samples were analyzed for TBA-reactive substances (TBARS) intriplicate, according to the method of Pfalzgraf and others (1995).
Meat samples (10 g) were mixed with 20 mL of 10% trichloroacetic acid in centrifuge tubes and vortexed for 2 min at high speed.
Samples were centrifuged (Jouan, model CR-4.11) at 2300 g for 30 minat 5 °C, and the supernatants were filtered through MN 640W filterpaper.
Two mL of the 20 mM TBA reagent were added to 2 mL of the filtrate,vortexed, placed in a boiling water bath for 20 min, and cooled.
Absorbance was measured at 532 nm.
The concentration of the samples was calculated using slope and intercept data of the calculated least-squares fit of the curve.
Results were reported as 2-thiobarbituric acid reactive substances (TBARS) in mg malondialdehyde/kg sample.
