2.4.2. Determination of cordycepin and adenosine
3′-Deoxyadenosine (cordycepin), adenosine, in samples of FCM
or DCM were analyzed by HPLC according to Guo et al. (1998) with
minor modifications. The HPLC analyses were conducted with a
Waters HPLC system consisting of a UV detector (2487), a pump
(A00515), an autosampling valve (7725i) equipped with a sample
loop of 20 μl used for sample injection, and Waters Empower
software for data analysis. The detection wavelength was set at
260 nm. The C18 column (Pinnaclell 5 μm, 250 4.6 mm) was a
Thermo Hypersil ODS. The optimum resolution was obtained by
using a linear isotonic elution. The mobile phase consisted of
disodium hydrogen phosphate buffer (pH 6.0) and methanol in the
ratio 85:15 (v/v). The flow rate was 1 ml/min.
