1. Introduction
Fluorinated molecules are becoming more and more important
in many areas going from bioorganic and medicinal chemistry to
agrochemicals and material sciences [1]. However the synthesis of
designed fluorinated compounds still remains a challenge in many
cases. From several years ago, we are developing an organic
synthesis programme based on the use of fluorinated propargylic
intermediates [2]. The main advantages of these derivatives
are their easy synthesis, including in optically pure form for the
monofluorinated compounds [3], and their versatility towards the
preparation of many new molecules including linear compounds
[4], carbocycles [5], and heterocyclic systems [6].
Various methods have been proposed for the preparation of
benzylic fluorides but in many cases there is still a strong need for
new and efficient strategies [7]. In connection with medicinal
chemistry programmes, we became interested in the preparation
of type A annulated aromatic derivatives with both mono- and
gemdifluorinated side chains and functional groups R1 in
appropriate positions. Towards this goal we designed a strategy
based on the use oftwo key steps as indicated in Scheme 1. A Diels–
Alder-aromatisation sequence will be employed to build the 6-
membered ring (through intermediate B), while intramolecular
enyne metathesis reactions starting from adequately substituted
propargylic fluorides D should afford the desired new dienes C
with the appropriate fluorinated side chains [8].
In this communication we demonstrate on two indanes,
selected as representative models, that this scheme is efficiently
working to obtain the desired target compounds. Scheme 2
2. Results and discussion
Our synthesis started from known [9] enyne 2, obtained from 1
[10] in 85% yield by metallation with n-BuLi and trapping by
acetaldehyde. Reaction with diethylaminosulfurtrifluoride (DAST)
in CH2Cl2 at 0 8C gave the desired propargylic fluoride 3 in 58%
yield. The intramolecular enyne metathesis reaction [11,12], using
Grubbs II catalyst at 5 mol%, proved to be very successful affording
the desired diene 4 in 91% yield. This intermediate was fully
characterised by spectral and analytical data. However it proved to
be labile and should be better used immediately after its
preparation. The Diels–Alder reaction with diethyl acetylenedicarboxylate,
neat at 50 8C during 24 h, gave the cycloadduct 5 as a
mixture of two diastereoisomers. These labile compounds were
neither purified nor separated and the mixture was used directly
for the next step. The aromatisation of this Diels–Alder adduct was
not straightforward since DDQ gave only degradation, while MnO2
gave only little transformation into the desired aromatic product 6.
Finally, this step proved to be best performed by using IBX in DMSO
at 80 8C during 24 h and the desired benzylic fluoride 6 was
