Chinese Spring (CS) is a low-frequency regeneration wheat
type with the embryogenic callus induction rate and plant
regeneration rate about the same of 0.37% with alone culture
(AC). In experiment 1, this genotype was inter-cultured with
Xinchun 9 (XC9), which is a high-frequency regeneration
type with 32.85% (AC) of embryogenic callus induction rate
and 41.61% (AC) of plant regeneration rate, respectively.
The results suggested the increased embryogenic callus
induction rate of 19.33% and plant regeneration rate of 18.59% for CS when inter-culture technique (IC) was subjected,
but for XC9 the embryogenic callus induction rate
and plant regeneration rate were almost the same between
AC and IC (AC: 32.85 and 41.61%; IC: 44.65 and 49.82%)
(Table 1, Fig. 1). In experiment 2, CS was inter-cultured
with another high-frequency regeneration wheat variety
Kenong 199 (KN199). The untreated embryogenic callus
induction rate and plant regeneration rate were 4.55 and
3.03% (AC) respectively in CS, and 74.66 and 176.82%
(AC) respectively in KN199. Inter-culture of these two wheat
lines resulted in a surprising increase in embryogenic callus
induction rate and plant regeneration rate. CS obtained
52.19 and 67.98% (IC), respectively. And the inter-cultured
KN199 reached similar rates of 78.29 and 151.16% (IC) to
those via AC (Table 1, Fig. 1).
