2.5. Antagonist bacterial isolates and in vitro inhibitory activity against isolated pathogen
In the present study a total of five bacterial strains were used to control isolated pathogens of which two were plant rhizospheric bacteria viz. Burkholderia cenocepacia VBC7 (NCBI GenBank Accession No: HM042678) and Pseudomonas poae VBK1 (NCBI GenBank Accession No: GU384238) (Mukhopadhyay, 2010) and another three different strains (LABW1, LABW3 and LABW4) of Lactococcus lactis subsp. lactis isolated in our laboratory and identified following 16S rDNA sequence homology. The two rhizobacterial strains B.cenocepacia VBC7 and P. poae VBK1 were isolated from the rhizospheric soil of Chick pea (Cicer arietinum L.) and mung bean (Vignaradiata (L.) R. Wilczek) respectively on nitrogen free Burk’s med-ium and were selected on the basis of their plant growth promoting properties. On the other hand all of the lactic acid bacterial strains were isolated from good quality fermented milk products (whey) on de Man Rogosa and Sharpe (MRS) medium and were selected for their very good antimicrobial activities. Antifungal activities of these five bacterial strains were checked against isolated pathogens by dual culture overlay assay (Magnusson and Schnürer, 2001). The antifungal activities of the cell free supernatants of the bacterial strains were also checked against isolated
pathogens by agar well diffusion method (Fernández-Garayzábal
et al., 1992). Here both boiled and unboiled cultures were tested.
Inhibition of percent spore germination of VBAM1 by different
dilutions of culture filtrates of five biocontrol organisms was tested
on cavity slides followed by microscopic observations (Saravanan
et al., 2004).
