-To determine soluble sugars in the stems of the 59 accessions,
2 g dry matter of each accession was added to 40 mL distilled
water and shaken at 150 rpm for 2 h at 50°C.
 -After centrifugation at 1800g for 10 min, the residue was washed five times with 20 mL
distilled water.
- As a result, the soluble sugars in the stem sample
were extracted into the supernatant. 
-The residue without soluble sugar (RWSS) was dried at 80 _C in an oven. Soluble sugar was
determined with the supernatants by the anthrone/H2SO4 method
and assayed in a UV–VIS spectrometer .
-The standard curve was plotted using D-glucose as a standard. 
-Dry matter (2 g of each)
was added to 30 mL ceramic crucibles to determine ash content
in a muffle furnace
