2.7. Transmission electron microscope
The autophagy of NIT-1 cells was evaluated by autophagosome
screening under transmission electron microscope (FEI Tecnai G212,
Netherlands). At the end of intervention, the cells were digested with
0.25% trypsin and collected in centrifuge tubes. Then the cells were
centrifuged at 1500 r/min for 10 min, followed by another centrifugation
at a speed of 800 r/min for the same time. The supernatant was
discarded and 2.5% glutaraldehyde was added into the tube to fix the
cells. After 15 min of fixation, dehydration, embedding, sectioning and
staining was performed under general supervision. Then the autophagosomes
were observed under a transmission electron microscope
and photographed.
