The growth medium used in the study included 1.5 L of potato dextrose
medium with 2% (v/v) of ethanol added before inoculation.
The initial pH was set at 5.6. The agitation was maintained at
120 rpm and the initial aeration rate was set at 1 vvm. The temperature
was maintained at 28 C through convective heat transfer
using a heating block containing a heating element and a cooling
coil. Spore inoculum used in the study was prepared as described
in Section 2.1.2. The inoculum was added to an extent that the concentration
of spores in the medium was 2 104 spores per ml on
day 0. Samples (in duplicate) were harvested every 24 h for residual
glucose and medium conductivity analysis. The biomass was
harvested from the reactor after 12 days (upon complete consumption
of glucose from the medium) for the estimation of biomass
and CPT production.
