• Metformin Extended-release tablets (Fortamet): Using single-composition osmotic technology (SCOT), metformin is released at a constant rate from an osmotically active tablet core surrounded by a semi-permeable membrane. Peak plasma concentrations are achieved at approximately 6 hours (range 3—10 hours). While peak concentrations are higher with Fortamet, the bioavailability of Fortamet 2000 mg once daily in the evening (as per AUC) is similar to the same total daily dose administered as regular-release tablets 1000 mg twice daily. When administered with food, the extent of absorption is increased by approximately 60%; in addition, Cmax is increased by 30% and Tmax is prolonged (6.1 hours vs. 4 hours). There is a dose-proportional increase in exposure following administration of 1000 to 2500 mg.
