2.5.5. High-performance liquid chromatography (HPLC)
The procyanidin profile of the Soxhlet ethanolic extract and
those of the 2nd step F-HPE and NF-HPE extracts were screened by
reversed-phase HPLC, following the method developed by Karonen
et al. [8], with some modifications. Samples were dissolved
in ethanol, microfiltered (0.20 m), vacuum dried and redissolved
in methanol to a final concentration of 40 mg/mL. The HPLC
system was equipped with a LiChrosorb RP18 column with precolumn
(250
×
4 mm i.d., 5 m), an UV detector (WellChrom k-2500)
and a HPLC pump (WellChrom Maxi-Star k-1000) from Knauer
(Germany). The binary mobile phase consisted of (A) water and
formic acid (99.5:0.5, v/v) and (B) acetonitrile. A linear gradient elution
was performed: 0 min: 100% A; 2 min: 100% A; 19 min: 91% A,
9% B; 35 min: 70% A, 30% B; 43 min: 30% A, 70% B. Column temperature
was 308 K, flow rate was 1 mL/min, detection wavelength was
280 nm and injection volume was 20 L. Epicatechin, catechin, caffeic
acid, gallic acid, quercetin, and taxifolin were injected in order
to try to identify these compounds in the extracts. This analysis was
performed in duplicate.
