2.1.1. Primary screening
All PNSB (earlier isolated from water and sediments from rice
paddy fields in Phatthalung, Nakhon Sri-Thammarat, and Songkhla
Provinces of Thailand) were screened to obtain isolates that grew
well in the presence of 0.25% NaCl (OD660 > 1.0), under both
microaerobic-light and aerobic-dark conditions. The concentration
of 0.25% NaCl in GA broth is equivalent to 4 mS cm1 electrical
conductivity, which matches the average of the saline paddy fields
in southern Thailand (Nunkaew et al., 2012).
2.1.1.1. Aerobic-dark conditions (Chemoorganotroph). 1 mL
inoculum was added to a screw cap test tube (150 25 mm:
50 mL) containing 9 mL GA medium with 0.25% NaCl (w/v), and all
culture tubes were incubated in a shaker at 150 rpm and 30 C, for
48 h. Dark culture conditions were ensured by covering the tubes
with aluminum foil, impenetrable by light. Bacterial growth was
measured using a spectrophotometer as previously described, and
