In most species, a strong stimulatory effect of Kp on gonadotropin secretion is unequivocal; the only exception is a report in fish showing an inhibitory role of this family of peptides [12]. Initially, evidence of the Kp stimulatory effect on gonadotropin release was obtained by injection into the cerebral ventricle of the mouse brain [8]. This finding was later widely confirmed in many species using intracerebroventricular, intravenous, or subcutaneous injections of Kp: rat [13] and [14], monkey [15], humans [16], sheep [17], pig [18], cow [19], mare [20], and goat [21].
The stimulatory effect of Kp on gonadotropin secretion is mostly mediated by a stimulatory action on GnRH release. Two lines of evidence point to this conclusion: (1) intracerebroventricular administration of Kp in sheep induced a large and sustained release of GnRH into the cerebrospinal fluid [17] and peripheral administration stimulated GnRH secretion into the hypophyseal portal system [22], and (2) the stimulatory effect on gonadotropin secretion has been found to be totally blocked in the presence of a GnRH antagonist [8]. The possibility that Kp could act also at the pituitary level has been suggested, but remains a matter of controversy. Kp has been shown to stimulate LH and FSH release from rat pituitary explants [14] and LH release from bovine and porcine pituitary cells [23]. In addition, dose-related LH and growth hormone secretory responses from dispersed pituitary rat [24] and bovine [25] cells were observed. In contrast, other studies have found no effect of Kp on pituitary response [13] or in altering GnRH-stimulated LH secretion in hypothalamo-pituitary-disconnected ewes [26]. By contrast, GPR54 is expressed in the human pituitary [27], Kp-immunoreactive fibers are located in the external zone of the median eminence in sheep [28], and Kp is released into the hypophyseal portal blood [26]. Hence, further studies are needed to clarify whether and how the pituitary contributes to the full gonadotropin response elicited by Kp.
