2.3. RecR selection process
The selection strategy for the RecR strain involved the exposure
of larvae from all generations to temephos concentrations corresponding
to LC for 50% (LC50) of larvae, estimated by dose–response
bioassays. For that, homogenous groups of 200 late 3rd instar larvae
(L3) were placed in plastic trays containing temephos in 1 L of distilled
water. After 24 h, mortality was registered and survivors were
transferred to clean water recipients. Larvae were fed until pupal
development, when they were counted and transferred to adult
cages. Live adults were also counted (by counting pupae exuviae)
in order to obtain the cumulative mortality until emergence.
