The most robust and generally applicable of preserving fungal strain is by freezing either in a mechanical freezer at –10 to –30 °C or in a liquid nitogen freezer at –176 to-196°C . Some species are readily stored in mechanical freezer at –10 toi –30°C after growth on agar slant
The strain in recoveried by thawing the slant and transferring the culture to fresh agar .this method has the obvious advantage of reduced handing and is effective over period of several years.it may not be applicable to all fungi and has a limited effective time for the recovery of viable culture , but in the short term ( 6-12 months) may be an attractive cost—effective machanism for preservation
In general fungal species can be stored in a liquid nitrogen freezer . Liquid nitrogen storage is generally accepted as the method of choice for preservation of physiological and morphological features .
