Conclusion
Mutagenesis under acidic stress seems to be a simple but effective method for improvement in vinegar production.
In this study, the wild type industrial vinegar producer – A. pasteurianus CICIM B7003 – was mutated by UV irradiation under acidic stress, and a mutant named A. pasteurianus CICIM B7003-02 evolved with higher tolerance, better productivity and more suitable characteristics for high-acidity rice wine vinegar fermentation. Maximum acetic acid accumulation for mutant is 103.81 1.17 g L-1 within 160 h cultured in Erlenmeyer flasks. Also, initial acid tolerance for the
mutant is increased by 33% than in the parental bacterium B7003. Further experiments revealed that the activities of ADH(s) and ALDH(s) in the mutant strain were 20–40% higher than those of the wild type. The value of this mutation not only is limited to the improvement in acetic acid bioproduction, but also will inspire the researchers who work on the microbial biosynthesis under other stress situations (such as alkaline, solvent and hyperosmotic agents).
