2.7. Analytical methods
To determine cell dry weight (CDW), cells were centrifuged at
10000g for 5 min at 4 C, washed twice with deionized water
and dried at 70 C to constant weight.
To determine the sugar concentration, samples were filtered
through 0.2 lm syringe filters, diluted if necessary, and then subjected
to high-performance liquid chromatography (HPLC) analysis
(Agilent 1200 series system, Agilent Technologies Inc.). Glucose,
xylose and other sugars were separated on an Aminex HPX-87P
column (Bio-Rad, USA) at 60 C using 5 mM sulfuric acid as the
mobile phase at a flow rate of 0.6 mL/min and a refractive index
detector (RID) (Zhang et al., 2012).
