2.3. STEC growth and inoculation
The STEC isolates were cultured independently in 20 ml Tryptic
Soy Broth (TSB) without dextrose (BD-Difco, Sparks, MD) using
50 ml sterile tubes at 37 C (150 rpm) for 18e24 h using a New
Brunswick Model G34 Environmental Shaker (New Brunswick,
Edison, NJ). In previous research we have found that use of TSB with
glucose alters the sensitivity of STEC by generating artificial acid
resistance not found in the actual food, which can lead to the isolates
appear more sensitive to the intervention technology (unpublished
data).
