Microbes from seawater and sediment
samples were serially diluted, whereas deep-sea
organism samples were homogenized and diluted
with artificial seawater (Hidaka and Sakai, 1968). An
aliquot of 0.1 ml of each of the samples was spread
on the medium. The colonies that developed on the
plates were isolated and stored at 3C after incubation
for 1 week at 27C.
