Count yeasts, using fluorescence microscope with blue excitation. Use 10X eyepiece with Howard mold count or
other eyepiece grid, and 20X (or 40X) objective. Count 3 squares of eyepiece grid in each field of filter not
covered by gasket. Count budding yeasts as 1 cell if daughter cell is obviously smaller than mother cell. If they
are approximately equal in size, count them as 2 cells. Count all yeasts located completely within an eyepiece
square and all yeasts touching left and lower border of eyepiece square. Do not count yeasts touching right and
upper borders.
