Explants of recalcitrant species may be highly discriminating for ionic strength of the
medium and therefore, to determine the best ionic strength, they should be subcultured three
to five times on different strengths of the same medium composition.We suggest the use of
different strengths (i.e., 5–20%, w/v) of mineral and organic content of seeds, with slight
modifications, for medium development. By applying this method, research on development
of culture media for other species is underway. If the applicability of this method can
be proven for other species, mineral and organic seed content may provide potentially a
universal approach for the development of a culture medium for most agronomic, forestry
and horticultural plant species. Since the seed compositions of many economically important
crops and nuts are already known, the method could dramatically shorten the time required
for the development of medium formulations for a large number of species.
