Biomass was separated by centrifugation (10 min, 3300 RCF)
and vacuum-dried to facilitate storage for further analysis.
HPLC analysis of saccharides. The filtered (0.22 lm) samples
(media- or post-cultivation effluents) were analysed by HPLC
(Waters 600S) using a Waters 717 autosampler and
300 7.8 mm Rezex Ca2 + column (Phenomenex) at 85 C. The column
was eluted with twice-distilled water at a flow rate of
0.8 cm3 min1
. Sugars were detected with a PrepELS II light scattering
detector (Gilson). Sugar concentrations in the reaction mixtures
were determined by external calibration of standard
saccharides and expressed as [g dm3
] (Mayer et al., 2004).
