2.2. Quantitative screening of phytase producing microorganisms
Five fungal isolates that made a zone of hydrolysis around its
colonies were inoculated in medium containing (g/100 ml):
Glucose, 1; KCl, 0.05; MgSO4, 0.5: NH4NO3 0.05; MnSO4,
0.01; CaCl2 0.5; FeSO4, 0.001; Yeast extract, 0.1 and Soya
been meal 10 g. The pH of the medium was adjusted to 7. Conical
flasks containing 50 ml of fermentation medium were sterilized
and inoculated with the different microorganisms and
incubated at 30 C in a rotary shaker adjusted to 200 rpm
for 48 h for five days. Phytase assay was carried out at the
end of incubation period.
