Using certain mods change your game so much that your current save file cannot run without those mods enabled. In cases like this when a new patch is out you will probably have to wait at least a day or more before the mods are updated.
If you simply hit "Play" instead of "Continue Campaign" you will be taken to the main menu where you can see that you are now able to play a new game without the mods. Also proving what i just said above.

I can't even get to the main menu. It goes to the initial starting screen (it says Rome II against the burgundy background) and just stays there. If I was to delete all my mods, play a campaign on CiG, and then when I see that the mods are updated again (mainly Divide et Impera) install them again, would it mess up my grand campaign? Basically uninstalling all of my mods, playing a new campaign on the DLC and then installing them all again when they're ready so I could continue where I left off on my grand campaign. Do you know if that would screw up my original save game?

Hello. I am also getting this problem, my game has been updated and removing all my mods doesn't seem to have worked either, I guess it is a problem that came with the update

I cannot play either, I think it's to do with opting into betas on the properties menu, but I cannot select it to opt in.

I had the same exact problem that was thankfully solved by the CA support team. This is copied from the email I got.
Please try all four of these steps before launching the game again and see if we can get you going:1. Run the Visual C++ redist setups again. They can be found here: C:Program Files (x86)SteamsteamappscommonTotal War Rome II
edist or your equivalent directory (varies according to OS)vcredist_x86-90.exe vcredist_x86-100-sp1.exe vcredist_x86-80-sp1.exe2. Rename the Rome 2 folder. This can be found by typing %appdata% in any Windows Explorer navigation bar then hitting enter. Navigate to The Creative Assembly folder and rename the Rome 2 folder.3. Verify the integrity of your game cache. Right click on Total War: Rome 2 in your Steam library and click Properties. Then navigate to the local files tab and click "Verify integrity of game cache...". This ensures your version of Rome 2 is exactly the same working version that appears on the server.4. Delete your Steam ClientRegistry.blob file. This can be found in your Steam installation folder (for me it's C:Program Files (x86)Steam)Please double check to make sure you have the latest system drivers, especially for your graphics and sound cards.

Side note with my experience. After running the game may create another Rome 2 folder. You'll know this when you start the game up and you have no saves. Just go back to the redist folder and delete the copied folder. Let me know if this helped!

Very helpful actually. I will say that after deleting all of my mods and reinstalling them my problem was fixed, but still you reminded me about cache integrity and updating drivers which will no doubt help performance. My gratitude.
