2.1.3 Paraffin Templates

Poly(vinyl alcohol) (PVA) (86.7–88.7% hydrolyzed, average molecular weight of 31,000) was purchased from Sigma Aldrich (Switzerland), and paraffin with a melting point of 56–58C was purchased from Panreac (Barcelona, Spain) and used as received. As previously described in the literature [25], for the preparation of paraffin microspheres a hot plate with magnetic stirring system was used as the heat source. A 1000mL glass bottle with 500mL water was heated to 60–70C. PVA (2.5g) was added with stirring to make a 0.5% w=v solution. 20g paraffin were heated to melt in a glass vial on the same hot plate. After adjusting the stirring speed, paraffin was added to the PVA solution. The mixture was then poured into a big beaker with 2000mL cold water to solidify the paraffin microspheres. Standard sieves (50, 100, and 355mm) were piled up from bottom to up in the previous order. The paraffin microspheres suspension was poured into the sieve tower. After the paraffin microspheres were rinsed with water to remove the residual PVA, they were separated further into two different size ranges: 100–355mm and higher than 355mm. Finally, paraf-fin microspheres were washed with distilled water five times and then lyophilized overnight. To prepare the paraffin tem-plates, plastic syringes (5mL) were chosen as the mould of the paraffin template. The end of the syringe was cut off. To obtain a paraffin template of about 3mm high, the piston was pulled to leave a gap of 3mm between the piston and the syringe end. Paraffin microspheres were transferred to fill the gap. The syringe end was pressed vertically on a piece of glass plate. The piston was pushed down gently to avoid deforming the paraffin microspheres in the process. The whole paraffin template was pushed out by the piston and stored into a desiccator.
