Regenerated and rooted plantlets of the fifth subculture were transferred to regular laboratory conditions with the container open for 1 day, and then rinsed in water to remove agar of the root before pot transfer. In order to evaluate the influence of different substrates and fungicide treatments, the plantlets were transplanted into pots containing sterilized perlite and others containing mixed of sand + compost (1:2, v/v), on the other hand one half of plantlets was treated with Pelt 44 (1 g/l) and the other with Benlate (1 g/l). The plantlets were grown in a controlled environmental chamber with continuously light. Each plantlet was covered with plastic bags in order to maintain a saturated humidity. Within 3 weeks, the plastic bags were removed gradually, and the plants were transferred to greenhouse under natural light condition, 70% RH and 25±2 ◦C.
