3.7. Kinetics
The Km and Vmax values of the partially purified amylase were
determined using different concentrations of soluble starch as
the substrate. The results are presented in Fig. 4b. The effect of
increasing substrate concentration on the enzyme reaction rate
follows a typical Michaelis–Menten equation with soluble starch
as a substrate. Double reciprocal plots using least square regression
to fit the plots were used to calculate the KM. It should be considered
that the lower Km value of starch is related to the higher
affinity for starch compared with other substrates. The Km and Vmax
values of the amylase enzyme were calculated as 2.7 mg/ml and
34.30 units/minute/mg of protein, respectively, at pH 5.0 and at
70 C (Fig. 4b). These results were very similar to those obtained
with the purified amylase enzyme from finger millet seed, which
had a km value of 2.8 mg/ml (Chethan et al., 2008). However, the
values are higher than those obtained by Kumari et al. (2010),
who reported that the km value of amylase was 1.25 mg/ml.
