The Secondary-conjugate (indirect) method requires the use of two primary antibodies generated in different species. Bethyl has prequalified primary antibodies for use in this form of the assay. Commercially available secondary-conjugated antibodies are anti-goat, anti-rabbit or anti-mouse (obtained by the researcher separately).
The Primary-conjugate (direct) method utilizes conjugation reagents (obtained by the researcher separately) and allows the use of primary antibodies made in the same species. This allows the researcher to choose from any of our primary antibodies, or to use an antibody of their own.
When identifying the necessary reagents for the proximity ligation assay, users will utilize either the SECONDARY-CONJUGATE (indirect) or PRIMARY-CONJUGATE (direct) pathway. See diagram below for the required reagents:
