Specimens of fish obtained for the development of this methodology
were authenticated by genetic analyses (sequencing). Also,
different species of bivalves, cephalopods, crustaceans, meats, vegetables
and spices were obtained to establish specificity of the
method (Table 1).
DNA extraction was performed for both methods (in-house
method and commercial kit) using the SureFood PREP Allergen kit (R-Biopharm) according to the protocol described by the manufacturer. The quality and concentration of extracted DNA was determine by measuring the absorbance at 260 nm and the 260/ 280 nm and 234/260 ratios using a NanoDrop ND-1000 spectrophotometer (ThermoScientific) (Winfrey, Rott, & Wortman, 1997). Subsequently, DNA extractions were labelled and preserved at 80 C.
