RNA Extraction and Reverse Transcriptase-PCR
Total RNA was isolated using a TRIzol reagent (Invitrogen
Co., Carlsbad, CA, U.S.A.) according to the manufacturer’s
instructions. Total RNA concentration was determined by
spectrophotometric analysis at 260 nm. In reverse transcription
reaction system, 2.5m g total RNA and 0.5m g random
primers were mixed and incubated at 70 °C for 5 min. Then,
5RT buffer, 20 UrRNasin ribonuclease inhibitor, 10mM
dNTP mix, 200U MMLV reverse transcriptase and RNase
free water were added. The mixture was incubated at 25 °C
for 10 min, 37 °C for 1 h and 95 °C for 5 min.
