shoots included)wereanalyzedusingthemethodof Graham
and Stevenson(1997). Aportionof3gofsproutstissuewas
added to100ml2.5%metaphosphoricacid,andhomogenatedfor
1 minatspeedsettingof5usinginahomogenizer(XL900,
Tianchuang, Changsha,China).Thehomogenatewascentrifuged
for 5minat5000g. A0.5mlaliquotofthesupernatantwas
then pipettedintoa10mlvolumetricflaskandfilledtoavolume
with 2.5%metaphosphoricacid.Theflaskwasshakentomix
thoroughly, filteredusinga0.45 mm nylonfilter(Millpore,
France), andplacedinanautosamplervial.Thechromatographic
system consistedoftheWatersmodel6000ASolventDelivery
System equippedwithaLDC/MiltonRoySpectroMonitorDUV
detector, Watersmodel717þ autosampler withachiller,andthe
Waters Millenniumintegrationsoftware.AZorbaxODS(250
4.6 mm)columnwasusedtoseparateascorbicacidwithamobile
phase of2%KH2PO4 (pH 2.4)ataflowrateof0.5ml/min.A10 ml
injection wasmadeforthestandardsandsamples.Foradeter-
mination ofthetotalascorbicacid,dehydroascorbicacidwas
reduced toascorbicacidusingthemethodof Hughes (1956).
Dehydroascorbicacidwascalculatedasthedifferencebetween
the totalascorbicacidandascorbicacidcontent.Resultswere
