2.1.3. Nutrient media
The 1/2 MS medium (Murashige and Skoog, 1962)
supplemented with 0.5mM naphthalene acetic acid, 0.3mM
GA3, 3.7 mM adenine sulfate, 10% coconut water, 0.5 g/l
activated charcoal, 80 g/l sucrose was used in all experiments
for testing the effects of culture substrate and culture system on
the formation of microtubers. Eight grams per liter agar was
added in control treatment in which explants were cultivated in
solid media.
The pH of media was adjusted to 5.8. In the control medium,
the pH was adjusted prior to the addition of agar. Media were
distributed inin vitro hydroponic system in aseptic condition
after autoclaving at 1218C, 1 atm for 30 min
