During 2012 thirty five batches (Table 1) of rHA were manufactured with the process described in the BLA for Flublok in the single 650 L stirred tank Bioreactor in Meriden, CT in order to make Drug Substance for the recombinant Influenza vaccine. Thirteen batches were H1 A/California, seven batches H3 A/Perth, thirteen batches H3 A/Victoria, and two batches B/Brisbane. The process is quite reproducible for individual strains. Variation between strains is more significant, for example HR/Victoria was almost three times more productive than H1/California. The same basic process (Universal Process) is used in all cases with allowance for changes in timing of infection and optimization of buffers and detergent for a new HA protein. The main change is the DNA sequence of the baculovirus. The Insect Cell Culture step is always the same.
