2.2.2. b-Glucan
AACC Method 32–23, AOAC Method 995.16 (K-BGLU 06/11,
Megazyme).
Samples are suspended and hydrated in a sodium-phosphate
buffer solution of pH 6.5 and then incubated with purified lichenase
enzyme. An aliquot of the filtrate is then hydrolyzed to completion
with purified b-glucosidase. The glucose produced is assayed
using a GOPOD, while absorbance is measured at 510 nm. Assay
kit is specific for mixed linkage [(1–3)(1–4)]-b-D-glucan. There is
a difference in quantity and factors when raw and cooked samples
are calculated.
