In order to determine the optimum pH of the crude enzyme
preparation from SSF, 0.2 M Glycine-HCl, 0.2 M
Acetate, 0.2 M Sodium phosphate and 0.2 M Tris–HCl
buffers were used in the pH range of 3.0, 4.0-5.0, 6.0-8.0
and 9.0 respectively. All the assays were done at a constant
temperature of 35°C.
