2.2. Sample and sample preparation
Cultivar Khao dok mali 105 was grown and harvested at a rice plantation in Kalasin, in northeastern region of Thailand in 2012.
After harvesting, the rice samples were cleaned up by removing
immature and damaged grains, which is the routine process of
industry. Then, 1.5 kg (sampling from one discreet spot from 15
places and mixing thoroughly) of rice sample was divided into
three subsamples. Five hundred grams was pre-germinated and
parboiled (Section 2.3) and dehulled, yielding parboiled germinated
brown rice (PGBR) and the remaining 1.0 kg was dehulled
to produce brown rice (BR). Five hundred grams of the dehulled
portion was further milled to obtain white rice (WR).
For chemical analysis, all three samples (PGBR, BR and WR) were
grinded with Cyclotec (FOSS, Sweden) and then sieved through 120
mesh sieve to obtain a homogenous fine flour. The flour was then
vacuum pack and stored at 18 C for further analysis.
