The total phenolic content (TPC) of the ethanol extract and its fractions was determined by the literature methods involving Folin–Ciocalteu reagent and gallic acid standard. 1 mL of sample was mixed with 0.5 mL of Folin–Ciocalteu reagent and 5 mL of distilled water. This mixture was kept for 5 min at room temperature. After that, 1 mL of 5% Na2CO3 was added and the mixture was incubated for 60 min in darkness at room temperature. Absorbance of the mixture was measured at 760 nm. The mean (±SD) results of triplicate analyses were expressed in terms of mg gallic acid equivalents (GAE)/g. The same procedure was repeated for all the gallic acid standard solutions (0–50 g/mL) and a standard curve was obtained.
