munosensor forthedetectionof S. aureus. Inthisbioassaysystem,abiotinylatedprimaryanti-S.aureus
aptamer wasimmobilizedonstreptavidincoatedmagneticbeads(MB),whichservesasacaptureprobe.
A secondaryanti-S.aureus aptamer wasconjugatedtosilvernanoparticles(Apt-AgNP)thatsensitively
reports thedetectionofthetarget.Inthepresenceoftargetbacterium,anApt/S.aureus/apt-AgNP
sandwich complexisformedontheMBsurfaceandtheelectrochemicalsignalofAgNPsfollowed
through anodicstrippingvoltammetry.Theproposedsandwichassaybenefits fromadvantageousofa
sandwich assayforincreasedspecificity,MBascarriersofaffinity ligandsforsolution-phaserecognition
and fastmagneticseparation,AgNPsforsignalamplification, andanelectrochemicalstrippingvoltam-
metry read-outasasimpleandsensitivedetection.Theelectrochemicalimmunosensorshowsanex-
tended dynamicrangefrom10to1106 cfu/mL withalowdetectionlimitof1.0cfu/mL(S/N¼3).
Furthermore,thepossibleinterferenceofotheranalogbacteriawasstudied.Toassessthegeneralap-
plicability ofthissensor,weinvestigatedthequantification of S. aureus in realwatersamples.Theresults
werecomparedtotheexperimentalresultsobtainedfromaplatecountingmethod,whichdemonstrated
an acceptableconsistency.
