2.4. The effect of callus age
To study the effect of callus age on callus growth and regenerability,
IEs of three inbred lines were initiated on callus induction medium
M11APC1 (Fig. 1a and b). Then, ten IE-derived calli from each line
were weighed at one, two, three and four week(s), respectively. To
study the effect of callus age on regenerability, six IE-derived calli
from each line were randomly sampled and subcultured on regeneration
medium B1I1C1 (Table 1) when they were one-, two-, three- and
four-week-old. After six weeks, regeneration rate and number of
regenerated shoots per IE were recorded individually.
