For the proliferation phase, embryos were carefully removed
from the explants and inoculated on semi-solid MS medium
containing various combinations of growth regulators. In most
of the cases callus was formed from these embryos. When
different concentrations of gelling agents were tested (Table 3),
10 g/L agar and 4 g/L phytagel induced formation of secondary
embryos along with callusing, but 12 g/L agar induced secondary
embryogenesis without callusing. Lower agar (8 g/L)/phytagel
(2 g/L) containing media induced callus whereas higher levels of
the gelling agent (agar—14 g/L and phytagel—6 g/L) resulted in
poor responses.
