Extraction and isolation of esculeoside A (1)
The fresh ripe fruits (1.78 kg) of Cherry tomato (Lycopersicon esculentum var. cerasiforme) purchased in Kumamoto city were smashed, added with water and centrifused. The supernatant was passed through Diaion HP-20 and eluted with water and MeOH, successively. The MeOH eluate was then subjected to ODS eluted with starting from 40% aq.
MeOH to MeOH, gradiently. The 70% eluate provided esculeoside A (1, 440 mg). Similarly, from the pink type tomato [L. esculentum (Momotaro)] (7.53 kg), esculeoside A (1) was obtained in a yield of 311 mg.
