Development and optimisation of an automated DNA extraction procedure
Initially, we isolated DNA by using the Maxwell 16 LEV Blood DNA Kit (AS1290) according to the manufacturer’s instructions, which resulted in a DNA yield below 10 ng lL-1 and an insufficient DNA purity. Therefore, we tried the same extraction protocol with maize seed and also with a different kit (Maxwell 16 Tissue LEV Total RNA puritifaction Kit, AS1220, Promega, Mannheim, Germany). The obtained DNA yield was slightly higher (AS1290[AS1220), but DNA purity was still insufficient. As CTAB buffer-based DNA extraction is most common and results in high yields, we wanted to combine a CTAB buffer extraction with the automated DNA extraction.
