Additional ingredients in regeneration medium have a significant
impact on regenerability. L-Asparagine and L-proline fostered significant
improvement in the number of shoots on B2C1AP (66.4 per IE) as compared
to that on B2C1 (56.3 per IE). PVP at 1 g/Lwhen supplemented in
B2C1P markedly increased shoot production (65.3 per IE) as compared
to B2C1 without PVP (56.3 per IE). These results are in agreement with
earlier reports (Elkonin et al., 1995; Hagio, 2002). A higher concentration
of CuSO4·5H2Oat1μM(ten-fold higher than that in MS) drastically
enhanced shoot production on B2C1 (56.3 per IE) as compared to that
on B2 (40.6 per IE), confirming the beneficial effect of high Cu level on
sorghum tissue culture (Nirwan and Kothari, 2003). Overall, B1I1C1
was the best regeneration medium for SA281 (Table 3).
