signal transmissionfromDpptoitstarget broad (br), agenewithacrucialfunctionineggshell
patterning.Weidentifyregulatorysequencesthataccountforexpressionofboth brk and br, andconnect
these tothetranscriptionfactorsofthepathway.WeshowthatDppdirectlyregulates brk transcription
through Smad-andSchnurri(Shn)-dependentrepression.BrkisepistatictoDppin br expressionand
activates br indirectly,throughremovalofarepressor,whichisyettobeidentified. Ourworkprovides
first cis-regulatoryinsightsintotranscriptionalinterpretationofBMPsignalingineggshellmorphogen-
esis anddefines atranscriptionalcascadethatconnectsDpptotargetgeneregulation.
