Overall cellulase (based on filter paper unit, FPU), endoglucanase
or CMCase (carboxymethyl cellulase), xylanase and b-glucosidase
activities were determined by method of Adsul et al. (2007),
without any modifications. Whatman No. 1 filter paper strips
(50 mg, 1 6 cm), CMC solution (1%, w/v), beechwood xylan solution
(1%, w/v) and p-nitrophenyl b-D-glucopyranoside solution
(0.1%, w/v) in 50 mM citrate buffer (pH 4.8) were used as
substrates for estimation of cellulase (FPU), CMCase, xylanase
and b-glucosidase activities, respectively. One unit (IU) of
enzyme activity was defined as the amount of enzyme required
to liberate 1 lmol/min of glucose, xylose or p-nitrophenol from
the appropriate substrates under the standard assay conditions.
Protein was estimated by Lowry’s method using bovine serum
albumin (BSA) as standard (Lowry et al., 1951).
