L-Asparaginase production was scaled up in a 30-L bioreactor
(Scigenics, India) with a working volume of 22.5 L. RSM-optimized
Czapek Dox Medium was inoculated with 2% (v/v) inoculum raised
in nutrient broth (O.D. 1.0) and fermentation was carried out at pH
6.0, 37 C and agitation of 200 rpm for 24 h. Air flow rate was
maintained at 0.75 vvm. Foaming was controlled by addition of
sterilized silicon antifoam. Samples were withdrawn every 2 h
for analysis.
