2. Materials and methods
2.1. Isolation and purification of polysaccharides
The dried fruiting bodies of Jinqian mushroom were pulverized by a miller. The power (100 g) went through a 60-mesh
screen and defatted with ethanol. The residues were dried at 45 ◦ C
and extracted with hot distilled water for 3 h at 90 ◦ C to obtain
water-soluble polysaccharide in yield of 11.34%. Subsequently, the
resulting materials were extracted with 5 L of 1.25 M NaOH/0.05%
NaBH4 solution twice at 25 ◦ C for 2 h. The combined supernatants
were neutralized by acetic acid, centrifuged to remove precipitation, concentrated and removed proteins by Sevag methods. Then,
the acetone was added slowly to the supernatants until the acetone
concentration (v/v) reached 50% volume. The precipitations were
obtained by centrifugation (8000 rpm/min at 50 ◦ C for 10 min),
thoroughly dialyzed (MW cutoff 3500) and lyophilized to give the
polysaccharide fraction JQPs (Fig. 1b, yield of 9.78%). A flowchart
describing the isolation of polysaccharide fraction JQPs was shown
in Fig. 2.
