2.5. Simultaneous sacchari®cation and fermentation
(SSF)
Pretreated and dry corn cobs (5±20% w/v) were added
to a 100 ml Erlenmeyer ¯ask in duplicate and autoclaved
for 15 min at 121°C and 15 lb/in:2. C. thermophile enzyme
broth (17 ml) was added to each ¯ask. Streptomycin
and Penicillin (10 mg/l) were used as bacterial
growth inhibitors. Tween-80 (0.04 ml) was added to
each ¯ask. Concentrated solutions of yeast extract and
peptone (2 ml each) were added to the enzyme broth to a
®nal concentration of 2% and 3% (v/v), respectively.
Finally, 1 ml yeast suspension of one day old culture was
added. The SSF was carried out at 37°C in a shaker at
150 rpm. Whole ¯asks were harvested after every 24 h
up to 96 h of incubation.
