3.2.2 Analysis of responses
3.2.2.1 Determination of brix content/Total Soluble Sugar
A hand held refractometer (OTANGO manual Model, Japan) was used to determine the total
soluble sugar (TSS) content. The refractometer was standardized by placing a drop of distilled
water on its prism. It was then positioned in a way that would allow entry of sunlight via the
prism. The eyepiece was used to observe the standardization after adjusting the coarse and fine
adjustments properly to a 0 % TSS. Subsequently, a drop of juice from a sample of MD2
pineapple was placed on the prism and the percentage (%) TSS determined. The process was
repeated for each sample and the percentage TSS determined (AOAC, 2000).
