The N-terminal amino-acid sequence of cellulase was determined with specimens electro-blotted to polyvinylidene difluoride membrane and ABI 492 protein sequencer (Applied Biosystems). The internal amino-acid sequences of cellulase were determined by mass spectrometry with tryptic and lysylendopeptidyl fragments prepared by the digestion with 1/200 (w/w) enzymes at 37°C for 12 h.
